Development of Inactivated Vaccine Against HFRS Using a Variant of Prototype Hantaan Virus Strain

Abstract

|
|
Haemorrhagic fever with renal syndrome (HFRS) is a serious and often fatal disease cause by viruses in the Hantavirus genus. The novel vaccine strain(CFC76-118) was selected from the prototype Hantaan virus 76-118 by serial passage in the brain
of
new
born mice. Inactivated HFRS vaccine was prepared from brain culture after treatment with formalin and purification by sucrose gradient ultracentrifugation. The characterization on the nucleocapsid protein and envelope proteins in the vaccine was
performed by SDS-PAGE, western blot and slot blot analysis. The amount of mouse myelin basic proteins and mouse serum proteins in the vaccine was measured by enzyme-linked immunosorbent assay(ELISA). Vaccine was in jected 3 times to guinea pigs
to
test
immunogenicity. The resulting sera were analysed by plaque reduction neujtralization test(PRNT) in vitro and virus challenge test in vivo. The results suggest that CFC 76-118 inactivated vaccine is effective against Hantaan virus infection.
